User Protocols
Here you will find a selection of user tested protocols optimized on different specimen and different requirements. Some of them are designed for lower throughput, but you can easily adapt them to a full tray by increasing incubation times. You can find more protocols like this in the InsituPro VS section.

Protocol of the Month December 2007
InsituPro UP Protocol Medaka
Configuration: 30 small sized baskets
Method: Double fluorescent in situ hybridisation
This protocol was established as a whole mount protocol for different developmental stages of Medaka embryos.

Template Protocols
Basically, you will find 2 protocols for IHC and ISH each for each configuration (small and medium baskets) and one clean protocol. The protocols are the same as for the InsituPro VS.
Protocol clean
Method: Sterilizing the tubings and baskets
This protocol should be used to clean up the incubation baskets prior to their first use. The baskets will be sterilized by rinsing them with 3% hydrogenperoxide at elevated temperature.

Template protocol ISH 1
Method: In situ hybridisation
This protocol can be used as a template for seamless adaptation of short manual in situ hybridization protocols. The template includes prehybridization and hybridization steps as well as the post hybridization washes, all performed at elevated temperature. The incubation with DIG antibody and intensive washing steps are included as well.

Template protocol ISH 2
Method: In situ hybridisation
This protocol can be used as a template for seamless adaptation of manual in situ hybridization protocols. It already includes most steps of a typical in situ hybridization method like rehydration of the specimen, bleaching with hydrogenperoxide, permeation using proteinase K and postfixation. The prehybridization and hybridization steps as well as the post hybridization washes are performed at elevated temperature.

Template protocol ICC 1
Method: Immunodetection
This protocol can be used as a template for seamless adaptation of short manual in situ immunostains. The template includes incubation with primary antibody and secondary antibody as well as all washing steps between and after these two incubations. Most steps are performed below ambient temperature.

Method: Immunodetection
This protocol can be used as a template for seamless adaptation of manual in situ immunostains. The template already includes most steps of a typical immunocytochemical method like rehydration of the specimen, permeation using proteinase K and postfixation. The incubation with primary antibody and secondary antibody as well as all washing steps between these two incubations, are performed at a temperature below ambient.
